Pulsatile Release

Pulsatile drug release is intended to deliver a burst of drug release at one or more predetermined time intervals. The need for pulsatile release may include the desire to avoid drug degradation in the stomach or first pass metabolism; the ability to administer two different drugs at the same time, yet released at different sites in the GI tract; or for chronotherapeutic drug delivery. Pulsatile release can be achieved via coating of multiparticulates with pH dependent and/or barrier membrane coating systems, followed by blending of the multiparticulates to achieve desired release profiles. As these multiparticulates spread throughout the GI tract, in-vivo profiles approximating to zero order result.
